Having the correct number of centrosomes is crucial for proper chromosome segregation during cell division and for the prevention of aneuploidy, a hallmark of many cancer cells. Like DNA, centrosomes are duplicated only once per cell cycle. Errors in centrosome duplication lead to cells with too many or too few centrosomes. The biochemical pathways underlying centriole duplication are largely unknown.
The aim of the project is to identify and characterize substrates of the master regulator of centrosome duplication, which is the polo-like kinase Plk4. Using biochemical techniques and yeast-two hybrid sceens we have recently identified a candidate substrate which is a so far uncharacterized protein. The PhD project will aim at characterizing this substrate in centrosome duplication and to analyze its regulation by Plk4.
Modern cell biological techniques including life cell imaging and high resolution confocal laser scanning microscopy as well as biochemical techniques will be used. The position will be for three years.
